Sodium Butyrate Supplement: Enhancing Animal Intestinal Function

Sodium butyrate, also known as sodium n-butyrate, is a short-chain volatile fatty acid whose core component is butyric acid. Sodium butyrate has gained prominence in the feed industry due to its unique advantages. As an antibiotic alternative, it has been widely accepted and applied in various types of feed within a few years, owing to its exceptional growth-promoting effects and safety.

Definition and Characteristics of Sodium Butyrate
Sodium butyrate is a short-chain volatile fatty acid that enhances digestive and absorptive functions in animal intestines. It has been widely used in various feeds. Its molecular formula is C₄H₇O₂Na, and it appears as a white powder with a distinct rancid cheese-like odor. It is highly soluble in water. Upon entering the animal intestine, sodium butyrate dissociates into sodium ions and butyrate ions, with the latter being the key active component. As a short-chain fatty acid, butyric acid is not only lipophilic but also remains stable in environments with a pH between 4 and 6.

Mechanism of Action of Sodium Butyrate
In the intestine, sodium butyrate dissociates into sodium ions and butyrate ions. The butyrate ions enhance intestinal absorption by stimulating villus growth and the activity of small peptide transporters. It is worth noting that the cecum and colon of animals naturally produce small amounts of butyric acid through microbial fermentation of sugars and proteins. Supplementing with exogenous sodium butyrate further enhances its ability to promote digestive and absorptive functions in the animal intestine.

Effects of Sodium Butyrate on Digestive Function
▲ Improved Digestibility
Studies have shown that adding sodium butyrate to the diet of Cherry Valley meat ducks significantly increased the digestibility of crude protein and crude fat (p < 0.05). Similarly, the apparent digestibility of dry matter and protein improved in Xiangyun crucian carp when coated sodium butyrate was added to their diet (p < 0.05). Additionally, calves fed with sodium butyrate exhibited significantly higher digestibility of dry matter, protein, crude fat, and calcium (p < 0.05). These findings indicate that sodium butyrate effectively enhances digestive efficiency in animals.

▲ Enhanced Digestive Enzyme Activity
Research by Chen Guoshun et al. demonstrated that adding coated sodium butyrate to piglet diets significantly increased amylase activity in the ileum and lipase activity in the jejunum by day 7 compared to the control group (p < 0.05). Wang Manman's study on Cherry Valley meat ducks found that when sodium butyrate was added at 0.035%, amylase activity in the duodenum significantly increased. At 0.07%, amylase activity in the duodenum, jejunum, and ileum was significantly higher than in the control group. These studies confirm that sodium butyrate significantly enhances amylase and lipase activity in the intestines of piglets and meat ducks.

Potential Pathways Affecting Digestive Function
▲ Regulation of Pancreatic Secretion
Sodium butyrate significantly increases pancreatic secretion by elevating Ca²⁺ concentration in acinar cells and stimulating insulin secretion. Studies injecting guinea pigs with different concentrations of butyric acid intravenously showed a significant increase in pancreatic secretion and amylase release, which escalated with higher butyric acid concentrations. Experiments confirmed that increased Ca²⁺ concentration in acinar cells promotes enzyme secretion, thereby stimulating pancreatic secretion.

▲ Role of Insulin
Increased insulin secretion helps stimulate pancreatic secretion, thereby enhancing digestive capacity. Studies found that intravenous injection of butyric acid in sheep significantly increased insulin levels in the intestine. Culturing rat RIN cell lines with butyric acid resulted in higher insulin mRNA levels in the experimental group compared to the control, further confirming that butyric acid boosts insulin secretion and promotes pancreatic secretion.

Effects of Sodium Butyrate on Absorptive Function
▲ Promotion of Intestinal Villus Growth
Multiple studies have shown that sodium butyrate significantly increases villus height and thickness, enhancing absorptive function. Research by Guilloteau et al. demonstrated that adding sodium butyrate to calf diets significantly increased villus height in the duodenum and jejunum, outperforming flavomycin.

▲ Increase in Villus Height-to-Crypt Depth Ratio
Sodium butyrate significantly improves the villus height-to-crypt depth ratio in broilers and meat ducks, enhancing absorption. Hu et al. found that while sodium butyrate did not significantly affect villus and crypt height in broilers, it linearly increased the villus height-to-crypt depth ratio.

▲ Regulation of Cell Proliferation
Experiments on piglets and broilers showed that sodium butyrate significantly increased the number of goblet cells in the colon, promoting cell proliferation. Studies indicated that adding sodium butyrate to piglet diets significantly increased goblet cell counts in the colon, even surpassing the effects of antibiotics.

▲ Modulation of PepT1 Activity
By stimulating CDX2 expression and insulin secretion, sodium butyrate upregulates PepT1 activity, promoting the absorption of peptides and amino acids in the intestine. Studies observed that adding butyric acid to drinking water significantly increased PepT1 mRNA expression in the colonic mucosa of mice, highlighting butyric acid's role in enhancing PepT1 expression. These findings demonstrate that sodium butyrate enhances digestive and absorptive capacity in animal intestines through multiple pathways, providing a scientific basis for its application in production.
