What is Albendazole Tablets?
Pharmacodynamics: Albendazole is a benzimidazole compound with broad-spectrum anthelmintic activity. Nematodes are highly susceptible, while strong effects against cestodes and trematodes also require higher dosages. It is ineffective against schistosomes. Its primary mechanism of action involves binding to tubulin in nematodes. Albendazole binds to β-tubulin, inhibiting its polymerization with α-tubulin to form microtubules. Microtubules are fundamental structural units of many organelles and are essential for cellular processes such as mitosis, protein assembly, and energy metabolism. Albendazole exhibits a significantly higher affinity for nematode tubulin than for mammalian tubulin, resulting in very low mammalian toxicity. This product is highly effective not only against adult parasites but also against immature worms and larvae, and it also possesses ovicidal activity.
Pharmacokinetics: Albendazole is one of the better-absorbed benzimidazoles after oral administration. Cattle absorb approximately 50% of the administered dose from the gastrointestinal tract. Within 9 days, 47% of the oral dose can be recovered in urine as drug metabolites. Following oral administration in sheep, the parent drug is undetectable or only briefly detectable in blood due to rapid metabolism to albendazole sulfoxide the primary active metabolite responsible for the anthelmintic effect. The metabolites albendazole sulfoxide and albendazole sulfone reach peak plasma concentrations around 20 hours post-administration. The elimination half-lives of the sulfoxide metabolite are approximately 20.5 hours in cattle, 7.7–9.0 hours in sheep, 5.9 hours in pigs, and 4.3 hours in chickens. The elimination half-lives of the sulfone metabolite are approximately 11.6 hours in cattle, 11.8 hours in sheep, 9.2 hours in pigs, and 2.5 hours in chickens. In addition to the sulfoxide and sulfone, other metabolites include hydroxylated, hydrolyzed, and conjugated products, which are excreted in the bile.
