What is Triclabendazole Tablets?
Pharmacodynamics: Triclabendazole is a benzimidazole anthelmintic specifically effective against Fasciola species. It demonstrates significant efficacy against all developmental stages of Fasciola hepatica, making it an ideal fasciolicide. After absorption, it disrupts the microtubule structure and function of the parasite and inhibits the release of proteolytic enzymes from the parasite. The effect of triclabendazole on the parasite is concentration-dependent: adult flukes survive for 24 hours at low concentrations (1–3 μg/ml); show significantly reduced activity at higher concentrations (10–25 μg/ml); and are completely inhibited at high concentrations (25–50 μg/ml). Immature flukes are more susceptible, with complete inhibition of activity occurring within 24 hours at a concentration of 10 μg/ml.
Pharmacokinetics: Triclabendazole has high bioavailability. In goats and sheep administered an oral dose of 10 mg/kg body weight, peak plasma concentrations of approximately 15 μg/ml are reached within 24–36 hours. The peak plasma levels of triclabendazole and its metabolites are 5–20 times higher than those of other benzimidazole anthelmintics, with an elimination half-life of about 22 hours. In sheep and rats, triclabendazole is largely oxidized to sulfoxide and sulfone derivatives. These derivatives bind to albumin and persist in the plasma for over 7 days. The high plasma concentrations and binding to plasma albumin appear to be associated with the prolonged activity against Fasciola species. In sheep, approximately 95% of the administered dose is excreted in feces within 10 days, 2% in urine, and less than 1% in milk.
