What is Furosemide Tablets?

Pharmacodynamics: Furosemide primarily acts on the medullary portion of the ascending limb of the loop of Henle, inhibiting the reabsorption of Cl⁻ and Na⁺. It also has effects on the cortical portion of the ascending limb. As a result, the concentration of Na⁺ and Cl⁻ in the tubular fluid increases, while the concentration of Na⁺ and Cl⁻ in the medullary interstitium decreases, leading to a reduction in the kidney's concentrating ability and consequently increased excretion of water, Na⁺, and Cl⁻. Due to decreased Na⁺ reabsorption, the elevated Na⁺ concentration in the distal tubule promotes increased Na⁺-K⁺ and Na⁺-H⁺ exchange, resulting in enhanced excretion of K⁺ and H⁺. Additionally, furosemide inhibits Na⁺ and Cl⁻ reabsorption in the proximal and distal convoluted tubules, further strengthening distal tubule Na⁺-K⁺ exchange and promoting K⁺ excretion.

 

Pharmacokinetics: It is readily absorbed after oral administration. In dogs, peak plasma concentrations are reached within 1–2 hours post-administration, with a bioavailability of approximately 77% and an elimination half-life of 1–1.5 hours. About 50%–60% of the dose is excreted unchanged in urine, while the remainder is conjugated with glucuronic acid. In cases of renal impairment, the plasma half-life is prolonged, and dosage adjustment is necessary during administration.

Side Effects

What are the potential side effects of Furosemide Tablets in different animal species?

(1) May induce electrolyte imbalances such as hyponatremia, hypokalemia, hypocalcemia, and hypomagnesemia. Additionally, azotemia may occur in dehydrated animals.

(2) May also cause gastrointestinal disturbances, anemia, leukopenia, and weakness.

What are the Precautions of Furosemide Tablets?

(1) Contraindicated in anuric animals. Use with caution in animals with electrolyte imbalances or hepatic impairment.

(2) Prolonged high-dose use may lead to hypokalemia, hyponatremia, hypocalcemia, hypomagnesemia, and dehydration. Potassium supplementation, combination or alternation with potassium-sparing diuretics is recommended, with regular monitoring of fluid and electrolyte balance.

(3) Concomitant use with aminoglycoside antibiotics and corticosteroids should be avoided.

(4) No withdrawal period is required.
